Preparation of anti-canine interleukin-31 receptor alpha polyclonal antibody and evaluation of its therapeutic effect in canine atopic dermatitis

Abstract Canine atopic dermatitis (CAD) is a prevalent genetically susceptible inflammatory and pruritic allergic skin condition affecting not only the health of dogs but also quality life their owners. Interleukin-31 (IL-31) interleukin-31 receptor alpha (IL-31RA) are essential for development pruritus in primates mice. Hence, it expected that inhibiting IL-31RA will be an effective approach to alleviate pruritus. The purpose study was produce anti-canine polyclonal antibodies (anti-IL-31RA pAbs) evaluate efficacy house dust mite (HDM)-evoked responses. Dogs were immunized with antigens formed by recombinant short peptides coupled BSA anti-IL-31RA pAbs. CAD model developed using HDM allergen stimulation, effects pAbs on reduction examined. Atopic Dermatitis Extent Severity Index (CADESI)-4 Visual Analog Scale (pVAS) utilized responses, tissue samples collected from inguinal area pathological assessment cell infiltration. results showed high titers (1:128,000) specificity effectively produced. In group, severity damage, score, infiltration level factors considerably elevated. Anti-IL-31RA relieved behavior compared placebo-treated dogs. conclusion, suppressed vivo anticipated novel treatment disorders such as CAD.
